      通过厌氧/好氧交替平板迭代法培养活性污泥,采用5-溴-4-氯-3-吲哚基-磷酸盐(BCIP)蓝白斑筛选法、 PHB 染色、异染颗粒染色进行高效聚磷菌的快速筛选,并基于16 S rDNA 对各菌株进行高通量测序和鉴定。 结果表明,4 株菌可以分解BCIP 产生蓝绿色菌落且厌氧PHB 和好氧异染颗粒染色为蓝黑色,除磷率分别为1 号菌85.51%,2 号菌91.06%,3 号菌95.51%,4 号菌89.13%。高通量测序结果表明,1 号菌为Exiguobacterium sp.ZWU0009(微小杆菌属),2 号菌为Bacillus sp. C10(芽孢杆菌属),3 号菌为Exiguobacteriumacetylicum strain SI17(乙酰杆菌属),4 号菌为Exiguobacteriumacetylicum strain BGSLP4(乙酰杆菌属)。

      The activated sludge was cultured by plate iteration method with alternating anaerobic and aerobic operation. Efficient phosphorus accumulating organisms were rapidly screened using 5-bromo-4-chloro-3-indolyl-phosphate (BCIP), dyeing of PHB and heterochromatic particles. Each strain was sequenced and identified by high-throughput sequencing based on 16S rDNA. The results showed that the four strains could decompose BCIP and produce blue-green. After staining, the PHB in the anaerobic stage and the heterodye particles in the aerobic stage were all blue black. The phosphorus removal rates of the four strains were 85.51% (No.1 strain),91.06% (No.2 strain),95.51% (No.3 strain),89.13% (No.4 strain) respectively. The results of high-throughput sequencing showed that the No. 1 strain was Exiguobacterium sp. ZWU0009,the No.2 strainwas Bacillus sp. C10,the No.3 strainwasExiguobacteriumacetylicum strain SI17,and the No.4 strainwasExiguobacteriumacetylicum strain BGSLP4.
